Jan Lesser
Manhattan Beach, Calif. / 55 years old / Lung cancer

The story:
Lung cancer, the type that killed TV anchorman Peter Jennings, is one of the nastiest tumors; few survive inoperable cases. But Jan Lesser did. In February of 2000, she was diagnosed with two tumors in her right lung, two more in her liver and another in her brain. In the next 10 months, three separate chemo regimens failed to halt the cancer’s rapid growth.

The Recovery
In late 2000, Lesser started taking an experimental drug called Iressa. After five months, her tumors had shriveled away. The drug only helped 10% of patients in trials. Today, Lesser still takes the drug every day and remains free of cancer. She has lived to see her three kids go to college and the birth of a grandson in 2006. But Iressa hasn’t done so well. In 2005, AstraZeneca stopped selling it to new patients in the U.S. after it failed to improve survival in trials pitting it against a placebo. Why did Iressa work for Jan? Nobody knows, but one reason may be that her tumor’s stem cells contained genetic mutations that made them exquisitely sensitive to Iressa, says Ronald Natale, Lesser’s oncologist. Finding out which mutations drive drug response could lead to a new generation of personalized medicines.
Barbara Bradfield
Puyallup, Wash. / 66 years old / Breast cancer

The story:
Barbara Bradfield’s breast cancer came roaring back in 1992, only two years after she’d had a double mastectomy. She had a marshmallow-size tumor on her neck and 16 spots in her lungs. She refused more heavy-duty chemotherapy and faced the fact she was going to die.

The Recovery
Then she found out she qualified for a trial of a new drug called Herceptin, which targets 25% of patients whose breast tumors have a certain mutation. She was one of the first patients to take it. Typically, it extends life by five months. But within six months of when she started Herceptin in 1992, Bradfield’s tumors had all melted away. They have never come back. Today, biotech researchers are trying to come up with improved antibody drugs that will give more patients spectacular responses like Bradfield had.
